Self-incompatibility (SI) refers to the phenomenon that a plant's male and female sex functions are normal, but it cannot be self-fertilized or fertilized by different pollen in the same strain. Self-incompatibility is an intra-species reproductive disorder widely present in flowering plants, which is important for species to avoid inbreeding and maintain genetic diversity. In the past decade or so, scientists have found that the increase in intracellular free Ca2 + in incompatible poppy pollen can stimulate self-incompatibility reactions. However, whether extracellular Ca2 + is involved in this process has not been confirmed. Recently, researchers have used non-invasive micro-measurement techniques to measure the Ca2 + flow in the poppy pollen tubes to illustrate this problem. Studies have found that Ca2 + influx and oscillation occur during pollen tube growth, and self-incompatibility protein (S protein: 14kD) can stimulate Ca2 + influx. This influx is located 50mm behind the tip of the pollen tube. These SI-induced Ca2 + influx indicates that the increase in intracellular Ca2 + in the SI reaction is caused by extracellular Ca2 + entering the cell. At the same time, it was found that the Ca2 + Channel inhibitors Gd3 + and La3 + inhibit the normal Ca2 + influx and oscillation of the pollen tube and inhibit the growth of the pollen tube. The non-damage micro-testing technique has unparalleled advantages for studying self-incompatibility. It can detect the oscillation signal of cells in vivo and dynamically, and can obtain a real and stable Ca2 + signal.
